My assumption is that power-ups wear off eventually on their own.
My guess is the sequel will introduce Wario. Not only is he a Contrasting Sequel Antagonist to Bowser, but the sooner they adapt him the sooner he can get his spinoff.
He is the third most iconic Mario antagonist after Bowser and Donkey Kong, both of whom were already introduced, so I think this is very likely.
So the effect of the Mini Mushroom on Bowser will also eventually wear off?

My personal theory is that Foreman Spike becomes Wario (with one of his workers forced by his boss to be "Waluigi"), in a bid to re-brand himself and his Wrecking Crew to ride the coattails of the Mario Bros.' newfound popularity as both handymen and heroes.
That would be an odd case of Composite Character. However, I also think Wario will be a human from Brooklyn that tries to run the coattails of the newfound fame and success of the Mario Bros, and Waluigi will be his (biologically unrelated) henchman.
I've said it before and I'll say it again: Danny DeVito would be perfect casting for Wario.

Bowser Jr. and Wario are both possibilities for antagonists in the next movie, which is interesting because we don't really have an origin story for either.

That doesn't seem likely. Nor do I see Wario and Waluigi being from the human world.

If they do introduce Daisy, I would want her actually be from Mario and Luigi’s world. Like have it so the Mario and Luigi are doing their plumbing business for Daisy. Maybe Luigi has a crush on her, and she ends up following them to Sarasaland. And then either she finds out that she was destined to be a princess there through some prophecy or she does something very miraculous for the Kingdom that she earns the title.
Would be hilarious if they reference live action Daisy

I think it would make sense that the second film involves Yoshi and Daisy, and that it ends with Mario returning to the mushroom kingdom, to see that it is taken over by Wario.
I bet John Leguizumo is just salty that he only starred in the crappy 1993 movie instead of this billion-dollar smash (which ended up being overtaken by Barbie).
